A single contamination event can void months of meticulous peptide research. Lyophilised powder purity gets most of the attention, but the diluent you reconstitute with is the real gatekeeper of experimental integrity — especially for multi-draw vials that sit in the fridge for weeks. Bacteriostatic water (BAC water) is the UK laboratory standard for that job. This guide covers what it is, how it differs from sterile water, the chemistry of the benzyl alcohol preservative, and the slow-drip protocol we recommend for reconstituting our research peptide range.

1. What bacteriostatic water is

Bacteriostatic water is a non-pyrogenic sterile preparation of USP-grade water for injection containing 0.9% benzyl alcohol as a preservative. It is designated for laboratory reconstitution of lyophilised research compounds where a single vial must be entered multiple times over the study period. Supplied in sealed borosilicate glass vials — chosen over soda-lime glass because it does not leach alkali or other impurities into the solvent over time.

Bacteriostatic vs bactericidal

Bactericidal agents actively destroy microbial cells; bacteriostatic agents inhibit replication. A "static" approach is preferred for peptide work because aggressive bactericidal chemistry can shift pH or trigger oxidative stress, degrading sensitive amino acid chains. Freezing the microbial population — rather than lysing it — protects the peptide sample without introducing reactive by-products that could skew analysis.

2. Bacteriostatic water vs sterile water for injection (SWFI)

These two solvents look identical in the vial and are frequently confused. They serve different protocols.

  • Sterile Water for Injection (SWFI) — single-dose only. No antimicrobial agent. Once the seal is breached, any unused liquid must be discarded.
  • Bacteriostatic water — engineered for multi-dose use. Supports repeated sterile entries over roughly 28 days from first puncture.

For high-purity research peptides such as Tirzepatide, Retatrutide or BPC-157, matching the diluent's longevity to the peptide's fridge stability is a cost-of-research decision: SWFI in a multi-draw vial routinely leads to total loss of the reconstituted peptide through microbial degradation.

Which solvent for which protocol

  • Single-session assay: SWFI is acceptable if the entire vial is used in one sitting.
  • Multi-draw study (days–weeks): bacteriostatic water is mandatory.
  • Peptides that will not fully dissolve in BAC water alone (AOD-9604, Ipamorelin, Tesamorelin): use a small volume of acetic acid water first, then top up with bacteriostatic water — full ratio is on the product page.

3. The role of 0.9% benzyl alcohol

Benzyl alcohol (C7H8O) is the aromatic alcohol that converts sterile water into a multi-draw reagent. At 0.9% it disrupts the lipid bilayer of microbial cell membranes, increasing permeability and halting replication. It freezes the microbial population rather than killing it outright — that "static" behaviour is the point.

The 0.9% concentration is a deliberate Goldilocks zone. Too low and robust strains are not suppressed; too high and the preservative can precipitate delicate peptide chains or introduce toxicity into research models. Benzyl alcohol also acts as a stabilising buffer, holding a consistent pH so peptides prone to deamidation or oxidation (e.g. Retatrutide, BPC-157) are not exposed to swings toward acidity or alkalinity in the vial.

Shelf life and the 28-day rule

  • Sealed vial: stable for up to ~24 months when stored cool and dark.
  • Opened vial: preservative efficacy is validated for 28 days from first puncture. After that window, discard — this rule is absolute in laboratory practice.
  • Storage temperature: 20–25°C for the diluent itself; the reconstituted peptide goes into the fridge at 2–8°C.

4. Slow-drip reconstitution protocol

Peptides are shear-sensitive. Blasting BAC water directly onto the lyophilised cake or shaking the vial can fracture the peptide chain, generate foam, and reduce yield. The slow-drip method protects the molecule:

  1. Swab the stoppers of both the peptide vial and the BAC water vial with 70% isopropyl alcohol; allow to dry.
  2. Draw the target volume of BAC water into a sterile syringe. Confirm the volume against the peptide reconstitution calculator for your target mg/mL.
  3. Angle the peptide vial and insert the needle so the tip touches the inside glass wall above the cake.
  4. Depress the plunger slowly — let the BAC water run down the wall onto the cake, not through it.
  5. Swirl gently — never shake. A slow rotation for 30–60 seconds fully dissolves the peptide without shearing.
  6. Label the vial with date of reconstitution and target concentration. Refrigerate at 2–8°C, protect from light.
