What is PT-141?
PT-141 is a cyclic heptapeptide that arose as an active metabolite of Melanotan 2. Where MT-2 has significant activity at MC1R — the receptor associated with pigmentation — PT-141 shifts the balance toward MC3R and MC4R, the central melanocortin receptors.
The melanocortin system
There are five melanocortin receptors. MC1R is associated with pigmentation, MC2R with adrenal signalling, and MC3R/MC4R with central nervous system pathways including appetite and arousal signalling in published research. PT-141's research profile is centred on the latter group.
PT-141 vs Melanotan 2
MT-2 is broadly active across the melanocortin receptors including strong MC1R engagement, which is why pigmentation dominates its research literature. PT-141 has comparatively reduced MC1R activity and is studied for the central receptors instead.
